Is very stiff; enough that a nickel will pop out of the screw head if you don't really bear down while adjusting.  It looks like the same staked on style of nut opposite the screw head that you get on S&W revolvers.  Lube is not helping.  Gun is 99% and no signs of abuse anywhere.  I need to free up the movement a bit.
Thoughts about cause and remedy?

I'm sure there are better answers, but the last time I had something like that, I put a small amount of penetrating oil on the gun, and left it overnight, so it could find its way to where it was needed.  I think that suggestion came from someone in this forum, but it was many years ago.  It did work for me.

One more thing - once you lubricate it with penetrating oil, do NOT try to turn the screw all the way in or out.  Just wiggle it back and forth, and once it starts to turn just a little wiggle it more, so each time you are turning it more.  Maybe add more penetrating oil.  Once it starts to move, it should/may eventually free up to where it works normally.  You don't want to break anything, so be gentle.

Here, Kroil is your friend.

As Mr. Meyers commented, just wiggle the screw forth and back just to allow the Kroil to work.

As you used a Nickel to move the screw, I suspect you have the 5.5" Heavy Barrel version in which the sight is identical to the Model 52 sight.

You might also lightly tap the screw with a small brass hammer, the key word is lightly.  This can brake any rust and allow the oil to penetrate.  Be very patient and take your time.

I suspect Kroil is just as good, if not better than penetrating oil, but I bought a small bottle of penetrating oil with a "needle dispenser" making it very easy to apply exactly where I wanted.

Yep, patience is key.  A small brass hammer with a punch is likely to help a lot, but don't "hit it", just give it a light tap.

I never tried this, but if you're using Kroil, maybe you could remove the barrel and dip it into a small cup of Kroil, and leave it overnight?

So, how long do your brake pads last?  Carbon fiber pads work well for heat dissipation, but do not do well in high wear situations.   lol!

Seriously though, good points on freeing the rear sight blade.   Wonder how well (or advisable) it might be to use an acid such as vinegar or even Coca-Cola on the threads of the screw in question?  Just a thought.
